¿Hay alguna manera de hacer un área rectangular 0 alfa (transparente) después de haberla dibujado?

He visto muchas preguntas sobre cómo dibujar un rectángulo semitransparente u otra forma en un lienzo después de haberlo dibujado. Mi problema es que quiero que un área rectangular sea alfa 0, pero parece que no dibuja nada encima de lo que hay allí. Entiendo por qué es así (estás dibujando 0 alfa encima de algo que ya está dibujado), pero quiero esos píxeles 0 alfa. Tiene que haber manera.

Básicamente, quiero definir un área que tenga 0 alfa después de que el lienzo ya tenga algo dibujado.
